Cryopreserved normal human epidermal melanocytes isolated from neonatal foreskin (HEMn, catalogue number C-002-5C, Lot number 3C0253) were thawed and plated into Medium 154 (M-154-500) supplemented with HMGS (S-002-5). At the end of the tertiary culture (2nd passage) the cells were subcultured into Medium 154 supplemented with either HMGS or the new HMGS-2 (S-016-5) growth supplement. Cells were plated in T25 flasks at a seeding density of 5,000 cells/cm2 in triplicate. At the end of each passage, the cells were harvested, the number of population doublings achieved calculated and the cell seeded into new flasks at the original seeding density.
